I am going through the CD measurement. The problem is the HT value goes higher than 1000 @ some particular concentration of my compound. I should dilute the sample so that HT value come down, but I want to study the CD spectra at that (higher) concentration only...is there any way to study the compound at higher concentration in CD????
When HT voltage goes beyond 600, the signal starts to become noisy. Beyond 700 or 800, the noise becomes disproportionate to the signal and the data will be completely useless.
HT voltage depends on some other things apart from the sample concentration such as the buffer or the solvent used to dissolve the sample. Make sure that the solvent does not have absorption in the wavelength range where you are recording the CD. You can also try reducing the pathlength of the cuvette. It is also desirable to keep the OD below 1 to get a good CD spectra.
Thanks Uttam Pal Sir....I am using cuvette of 2mm. The solvent used is not absorbing. But what is OD? I haven't noticed it before in the instrument... How I can control it??
OD is the optical density. You should measure it in a absorption spectrometer. Also check the nitrogen level while recording CD. Oxygen absorbs in far-UV, which also interferes with the CD spectra. Chloride salts in the solvent, DTT etc. are also bad for CD. So, check that the compound is pure. The solvent may not have CD signal but check the HT contribution of the solvent.
