I'm growing hESC cells that are recovering from a transfection in a 6 well plate. They form colonies but I find it quite difficult to pick the colonies. When I detach them using a colony picking tip, they detach as a layer but won't get picked up by the tip. I've also tried another method to wash the cells with DPBS and aspirate all the liquid and to work quickly to pick up the colonies before the DPBS crystallizes (removing the liquid prevents the detached colonies from moving around as I try to pick them up). I'm new to this and I am wondering if there is a more sophisticated way to do hESC colony picking? Thanks in advance for any help and suggestions!
