I added 18.2 ml of chloroform in 1 g of tocopherol acetate. I tried to dissolve the mixture in cell culture media but it is separating into 2 phases. I tried adding ethanol, DMSO and more chloroform in varying concentrations. I want to mix tocopherol acetate in media such that there are no loses during filtration process.

The solvent like chloroform is an organic solvent that is insoluble in water, and two distinct liquid phases, an organic phase and a water phase is formed when they are mixed together. In the chloroform-water mixture, the water phase is on the top and the chloroform organic phase will be at the bottom.
Tocopherol acetate is also freely soluble in ethanol. So, I suggest you use ethanol as a solvent to dissolve tocopherol acetate. The final concentration of ethanol in culture should be less than 1% in order to avoid toxicity. Always include a vehicle control in your assay.
