I have tried many things like, took medium supernatant and added with hexane, ethyl acetate, chloroform but none of these are able to extract antimicrobial compound from supernatant..

I also did polarity based extraction, kept medium supernatant mixed with ethanol (1:2 ratio)and left it overnight at -20*c, then centrifuged this and took pellet, resuspended it into hexane and centrifuged again, took hexane and dried it in speed vacuum machine, and added 2% DMSO solution into dried eppendorf..Further i added ethyl acetate into remaining pellet and did the same as done with hexane...at last i added water into pellet and dissolved the pellet...But none of them is showing inhibitory activity as it was showing earlier. What can i do to extract antimicrobial compound..
