I am very much agree with Mr.Bocancea Igor, to minimize the error and to make your work question less you should always use 5 to 6 replicates of injections. There are lot of well established references available in support of my and Mr.Bocancea Igor’s statement.
Yes Amir, it is enough to inject 6 (or 5 minimum) time to calculate the RSD for the solution of System Suitability (which is usually the standard, for the assay... ) and it is not necessary to inject for 6 time each, all the samples (standard, probe, reference etc.), it is enough for 3 injection.
Pawel, the RSD of the peak area, which is calculated from several injection is dependent less of response of the detector (only if your peak height is almost the height of noise) but is dependent of pumping system (i.e.pressure), the mode of injection (e.g. manual or autosampler), the type of the solvent of the samplers, which can be different from mobile phase, the unstability of the mobile phase which contain ion-pair reagents etc.
Yes , RSD can be calculated on three injection.% RSD limit we can estbalish based on the devlopment experience. For low concentration standard we can not give 2% RSD for three injection For eg. diluted standard in related substances test.
