I am fractionating a protein hydrolyzate (enzymatic) obtained from a vegetable protein concentrate. I want to obtain fractions of 10Ka, 5 KDa and 3KDa using Amicon Ultra 15 filter. Previously the enzymatic hydrolysate was freeze-dried. I need to resuspended the for added that suspension to each tube (10KDa MWCO) according to the manufacturer's recommendations, and with the permeate pass it through another 3KDa tube. Do you have any idea what must be the usual initial concentration should be to maximize recovery? .
Few works mention the concentration of protein or sample for ultrafiltration.
I´m trying 20 mg/mL but i don´t know if it is appropiate.
