I want to know about the methods of outer membrane disruption without breaking the cell integrity. I know about the French pressure cell method, but I want to use another one. So that I can isolate the outer membrane proteins only.
If you just want to avoid the French press but it doesn't really matter whether the cells are disrupted or not, you can use a sonicator. The total membrane fraction is prepared by high-speed centrifugation. If you then want to isolate outer membranes, you can use the N-lauroylsarcosine (Sarkosyl) method, which dissolves the inner membranes, leaving the outer membranes intact. The outer membranes can then be purified by high-speed centrifugation.
There are many descriptions of techniques for outer membrane purification in the literature. The precise protocol will depend to some extent on the species of bacteria. Here are a few papers:
