I would like to introduce one of the important functions of ACK.
As shown in the below scheme, ACK (activated Cdc42-associated tyrosine kinase) also referred to as Tnk2 belongs to the ubiquitin-binding protein family and requires Nedd4-1 as E-3 ubiquitin ligase. These molecules contribute to the ubiquitin-dependent proteasome protein degradation pathway.
Mutant ACK resistant to the ubiquitin modification by ACK results in the reduction of the EGFR degradation, which is why ACK is related to the aberrant EGFR proliferative signaling in non-small cell lung cancer.
