How can we make a good lysis buffer for the skin tissue for the western blotting and what should its composition be? Is there any particular protocol for Western blot analysis of skin tissues?
The RIPA lysis buffer described by Abcam for their skin lysate is widely used: https://www.abcam.com/mouse-skin-tissue-lysate-total-protein-ab4025.html
I tried Ripa before and it worked very fine, first you have to cut the biobsy into very small pieces, freeze it with liquid nitrogen, grind smoothly, and then apply the lysis protocol by RIPA and PBS. You need to perform all steps on ice.
