I have green micro-algae (Enteromorpha) powder and want to measure the carbohydrates and proteins before and after the anaerobic digestion of this micro-algae with anaerobic sludge. Please suggest some methods to measure these two.

I am not sure about measurement of carbohydrates, but for proteins you can try conventional Kjeldahl method (automator would be helpful).

For carbohydrates you may use the method and analyzer developed by Gerhardt. Fibertherm is a very reliable equipment for determination of cellulose, hemicellulose and lignin in biomass samples. http://www.gerhardt.de/en/analysis-methods/plant-fibre-determination/

For protein quantification you can use Lowry's method. I am applying Lowry procedure to my microalgae cultures, because at this wavelenght their photosynthetic pigments do not absorb (734 nm).

This chromatographical and spectrophotometrical method is maybe more suitable to your needs, I don't know which infrastructure you have available. Good luck!
