I am working on protein expression by Pichia pastoris. I focus on mixed feed to get higher result.
I read many papers. Some of author said that glycerol is repression the AOX1 promoter, which is active under appearance of methanol, and sorbitol is the good substrate to take the higher protein expression yield. It means, when they use glycerol to mix with methanol in induction phase even just a little bit of glycerol proportion, the target protein is decrease. Some said that glycerol could improve the result and sorbitol is not good enough.
I know that these result depend on their own yeast strain. But there are some author use the same strain but the have the different result. So, I really want to know what is the specific different in these strain make the different result.
Could you help me with an idea for this question? I will try to figure out from your suggestion.
