I am working with P6 and P30 rats and I need a loading control to compare and normalize these samples. I have tested actin, tubulin and GAPDH but none of them is constantly expressed between these two stages. Any suggestions?
For different stages of development or treatments with global effects I recommend a total protein control such as amidoblack staining once you have finished blotting your membranes.
I am not an expert on skeletal muscle, but I would try some thing like myosin, a muscle specific collagen, or some nuclear antigen like nuclear pore complex protein, for which excellent antibodies are available.
