When I over expressed an E3 ligase Cbl mutant in a 3T3 cell line, the cell morphology changed. I have repeated this several times. Is this related to transformation? The cells are selected for stably expressing Cbl.

I am testing if they are really transformed in a soft agar assay now.

some more information and answers to some of the questions raised here

1) this Cbl mutant is a dominant negative E3 ligase as some point out

2) the morphology phenotype seem to attenuate after several passages, though I was not able to see a noticeable expression level change of Cbl during this change.

3) when co-expressed with a RTK, the phenotype goes more dramatically

4) control stable cell line with a empty vector is normal

5) I had two independently made cell line, and both show the same morphology phenotype shown here

6) after trypsinization, the cell size are same between WT and mutant cbl

7) I started with same number of cells when looked at the phenotype.
