I am needing to run a couple of assays that will quantify oxidative or alkylative DNA damage in vitro and in vivo of bacteria. I will be using a lab synthesized compound from my mentors previous research to apply to the DNA which will then be photoactivated by UV-A. There is a proposed mechanism for this reaction which produces DNA damage by either guanine oxidation, base alkylation, or hydrogen abstraction.
Due to the limitations of our laboratory, we cannot conduct any type of radioactive assays. Is there an assay as well that can provide a mechanism for the reaction occuring?
