What is the time course required to perform DNA damage by hydroxyurea for gene expression study?

I have grown plants in MS media and I want to do gene expression study during DNA damage by hydroxyurea. Suggest me the time courses required for DNA damage by hydoxyurea treatment in plants for gene expression study. I am also required the suitable methodology for analyzing DNA damage in plants which treated with hydroxyurea.

In animal cells, HU treatment is typically carried out for 16 hours followed by 8 hours of recovery. Not sure about plants but this paper has used 2.5mM HU for 24 hours. Hope this helps.
