Please tell me the terminology of the chromatography column which is made manually. I mean, I have to purify of protein in larger scale. I will make a chromatography column by myself, but I do not know which word that is used to express this column in the science papers.
One more question. My column is cation exchange column. I know that I must test the column before I use it to do the experiments. I just only think of the test for pressure. Do you have any ideas?

Resolution, HETP (see chromacademy.com)... You must qualify the column as all column manufacturers do. Why did you make your own column rather than purchasing a commercial analytical column? How do you expect reviewers to review your article if they can't make your column?

On the issue of terminology, I would suggest that you include the following specifications:-
- The composition of outer and inner column walls.
- The dimensions of the column inner and outer length and internal diameter.
- The composition of the column packing. If you used a slurry, to initially pack the column, I do not think that it is necessary to mention the solvent of the initial slurry. However, you should mention the composition of the final packing solvent.
- The particle size of the packing material.
Will the column function correctly with a range of mobile phases? Does the column have restrictions on the ranges of pHs and temperatures, which may be used?
What will cause the column to deteriorate?

I think the terminology you are looking for is "self-packed" column. Regarding how to test if a self-packed column is working properly or not, there are lots of published material. Please google "how to characterize self packed columns" and I hope you will find a set of criteria suitable for you. Good luck!

Your question gave me a good laugh. If you are old enough (old as dirt, for example) you remember (as I do) when we used to note in our papers that the column was a commercially-available column! It was assumed that you packed your own columns, so you noted if you hadn't.

You are asking how to let people know that you packed the column. I am pointing out that in the past it was the opposite. We used to note when the column was NOT hand packed, not vice versa. The assumption now is that the column is commercial, whereas before the assumption was that the column was made in the laboratory.
