I know they are found in microsomal fractions of endothelial cells, suggestive of an ER distribution, but why has their subcellular distribution not been visualized, e.g. IHC and confocal, in native tissue? Is it a methodical limitation, unreliable antibodies perhaps? I have never seen one publication demonstrating this.

I am not an expert in sub cellular localizations of your type of proteins but I wonder if you will find some valuable information in the massive amount of data available in the "Human Atlas Project" data base. You may already have seen the recent presentation of the type of information freely available in the research article "Tissue-based map of the human proteome" by Uhlen et al. Published in "Science" on January 23 this year.

I will have a look at the above. I have heard somewhere before that the lack of specific antibodies at this time precludes their detection in subcellular domains. Someone else told me that in the 90s they had unpublished observations that there was greater ablumenal release of EETs than lumenal, which would appear consistent with my hypothesis (well, gut feeling) that in resistance vessels CYPs may be localized within trans-IEL endothelial cell projections, which are putative signalling microdomains containing various vessel-specific combinations of TRP and KCa channels, ER/IP3R (discrete pools), connexins and Noxs. It has also been demonstrated that these regions may be able to locally translate their own proteins.
