I'm planning to use EpiTect Whole Bisulfitome kit from Qiagen to amplify bis-DNA.
ctDNA was isolated from plasma and I don't have recommended 50ng of converted bis-DNA for the amplification, would the assay work with less? Has anyone tried?
Have not tried the experiment but with many kits the entire reaction can be scaled down. The reaction seems to include basic buffer components and a DNA polymerase, I would expect the enzymes activity is independent of it's volume, in fact the limiting factor working with enzymes is often how little you can pipette! I have scaled down kit reactions by a factor of 4 with the only repercussion that yields are lower, as would be expected. The Quiagen kit says it should result in a 50 fold increase in your DNA yields so that's what you should get out of whatever you put in. I'm sure you could scale it down and try the reaction with 10ng bis-DNA, 0.2µL polymerase, and 5.8µL buffer.
