The virus I'm working with is a morbillivirus. I have approximately 50mL of cell culture media and want to get the virus as concentrated as possible to infect primary canine PBMCs. Thanks in advance for any help.

I've used ultracentrifugation to get the highest total yield.
But LentiX (PEG based concentrator) also works reasonably well and take less time and no special equipment. Good luck!
Thanks Adam. Trying to avoid ultracentrifugation. Planning to use a centrifugal filter similar to one of the articles you provided.
Depends on the virus (particularly its fragility). Most direct and simple way is moderate speed centrifugation.
You can use Clontech Lentivirus Concentration—Concentrate Lentiviral Supernatants Without Ultracentrifugation. Lenti-X Concentrator provides a fast, simple, and highly efficient method for concentrating any lentiviral stock. In the simple protocol, you just mix your lentiviral supernatant with the lentivirus concentration reagent, incubate for a short period, and spin the mixture in a standard centrifuge
