As per my knowledge the Topoisomerase helps in the unwinding of the double helical structure of the DNA, during replication as well as transcription. In which it binds to the dsDNA and cuts the phosphate bond , either on one or both the strands and helps the DNA to unwound. This is generally an isomerase enzyme that works on the topology of the DNA.
