We have cell cultures from tet-on animals, so they have been on tetracycline to keep the GOIs off for all their lives. What would be the correct does of tetracycline to apply to these cell cultures? Any direction/answer will be much appreciated.

I use cells expressing the tetracycline repressor and treat my cells with doxycycline 100 ng/ml

if it is a comercial or common used cell line, you can search for papers reporting tetracycline concentrations. If you are suspicious about antibiotic toxicity or not enough selection power, then you can perform a cell viability test to determine the concentration for your cells
