Due to the deleterious mutation, the stability of the protein may be decrease and DG will be negative. What is that stability means?
According to the Gibbs free energy formula (ΔG = ΔH - TΔS), ΔS is the change in ligand flexibility. Mutations usually increasing ΔS and therefore decrease ΔG and therefore decrease protein stability at the mutation point.

(or a silent or a non functional protein). I suppose you mean disease. However if you mean stability... Maybe in biological terms is something you would not need. Now it comes to placing this protein/organism in a different place, a suitable one. Now you need to ask about a beneficial mutation.
