I am planning to do scRNAseq on whole lung tissue. My aim is to achieve a compromise between keeping immune cells vital and digesting solid tissue to single cell suspension.
I stumbled about many protocols. Especially the cold-active Protease Subtisilin A sounds very interesting for my purpose. One comparison was made by Cohen (2019, figure attached).
Now to my question. Did any of you have any experience regarding Lung digestion for single cell analysis? My aim is to get a broad spectrum of immune, epithelial and endothelial cells. I dont need to sort before sequencing.
