How much the cells that I need to use for developing growth curve of the cells? What is the size of cell culture plate that I need to use? Is there any standard guideline to follow? At the moment, I used RAW 264.7 cells and synoviate cells. Thanks.
You can use 6 well plate for this study. Maximum number of RAW264.7 cells per well in a 6 well plate can be accommodated is 2 X 106 . Minimum number of cells that can be taken will be 50,000 ; so that cells will not grow in colony. Area of 6 well plate will be sufficient to accommodate the cells to be produced in 6 generations, if you are taking 50,000 cells to begin with.

The density, in which the cells are seeded, can affect their growth. That is why I recommend you to use 2-3 different cell concentrations. If you want to fix the optimal conditions for culturing these cells and to understand better their growth characteristics, you can also check the influence of some other parameters - media, sera, even different plating surface, etc. It is always helpful and will allow you to improve your personal experience.
