I need to immobilize 3 peptides: CGPDGRDGRDGRDGR, CRQILIWFQNRRMKWKKYQRLC, penetratin peptide RQIKIWFQNRRMKWKKGG on the SiO2-covered gold nanoparticles. What is the most efficient and stable way to do that? Is it better to modify proteins somehow? I do not have much experience in protein chemistry, I am spectroscopist.
Why don't you use plain gold particles? Two of your peptides contain cysteines, which would strongly and spontaneously bind to gold. To keep the immobilization consistent, I would order the third peptide also with a terminal cysteine, e.g. RQIKIWFQNRRMKWKKGGC.
I want to lower cytotoxicity and increase stability so I prefer silica shell on gold
In this case, you may modify the particles with an epoxy silane and conjugate the cysteines of the peptides to it.
