Hello everyone, in our lab we want to create lymphoblastoid cell lines for genetic studies. I have no experience in this field, I have read a lot of literature, but I am a bit confused. so I have some questions for those who would like to help me and are familiar with this field:
1) Some talk about separating the blood components by centrifugation, others argue that it is not necessary because EBV binds exclusively to CD21 possessed only by B lymphocytes.
2) To infect B cells, some use feeder cell cultures (MRC-5 cells) in which to propagate the viral component. Then, they take the supernatant (with virions) and use it to infect lymphocytes. Others, however, use marmoset B95-8 suspension cell culture to prepare viral broth and then use it directly to infect lymphocytes.
