I am not sure, what is your source of cytokines, but if it's cell supernatant, you do not need any protease inhibitor. You can directly detect it using commercially available Elisa kit

I use the minicomplete tablets from Roche when I homogenize tissue for assaying.  They are convenient and seem to work well.

I am planning to use multiplex Cytokine Kit for assay. If I need protease inhibitor during homogenization, is it better to use compete tablet or EDTA free tablet? 

I am not familiar with the EDTA free product.  I shouldn't think it matters for tissue, but you can check with the kit manufacturer.
