I need a good simple and easy protocol for measurement of ion leakage from plant root cells after heavy metal treatment. How can I measure the exact amount i.e. % of ions leakage ( need the formula) against a standard.
To know the amount of solute a you can makes a standard curve with KCl, it probably what is recommended with the booklet that comes with your conductivimeter. Once you have measured the conductivity of all of your sample it's good to boil all your sample for 10 minutes that gives the total amount of solutes in your sample and may account for small sample errors like different sample size. The assay its self is easy, you just float your leaf or piece of leaf over ddwater for 1/2 hour.
