I want to estimate the cholesterol levels in different tissues like adrenal gland, ovary, liver etc. Can anyone provide me the method of estimation of cholesterol in these tissues.
For liver samples and liver liver plasma membrane cholesterol determinations in many of our studies (Al-Othman et al. Proc Soc Exp Biol Med 204: 97- 103, 1993) ( Lei et al. Proc Soc Exp Biol Med 199: 335- 341, 1988), we have used the Folch et al. (J iol Chem 226:497 -509, 1957) method for total lipid extraction and the Roeschlau et al (J Clin Chem Clin Biochem 12:403- 407, 1974) for cholesterol determinations. David K. Y. Lei.
In my research first I extract lipids according to Blygh and Dyer (1959) and next I apply the method of Sperry and Webb (Sperry, W. W., M. Webb, 1950. A revision of Schoenheimer-Sperry method for cholesterol determination. J. Biol. Chem., 187: 97-101.) for determination of total cholesterol content. This I apply for muscles and plasma .
