The classic paper for ascertaining SMC in vivo migration was published in 1986 (in rats - but the principle is the same). Clowes and Clowes, Circ Res 58: 839-845, 1986. This involves the use of tritiated thymidine, but can be updated for BrdU etc.
One way I read in a paper is you physically scrape cells off a tissue culture plate (like with a blade or something) and observe rate of migration into the portion from where the cells were scraped off.
