I am using carboxy-H2DCFDA to detect ROS in Saccharomyces cerevisiae. I will load the dye before incubating my cells with the desired agonist. What is the desired time period till when I can detect the fluorescence of the DCF produced?
Forty five minutes to one hour should be ideal. Keep in mind that dead and dying cells produce ROS and you may want to exclude them from your analysis with the live/dead stain of your choice.
Please, consider Chen, X etal, Free Radic Res. 2010 Jun;44(6):587-604 for discussion of specificity of DCF-reaction, to my knowledge it does not detect ROS in general rather than hydrogen peroxide
Normally with the test DCFDA, it is possible to treat the Saccharomyces cerevisiae with the molecule of interest and the DCFDA simultaneously then incubating these cells for 3 hours at 30°C. After this incubation the measure of the fluorescence intensity is possible.
