I'll work with DMSO as solvent for dissolving my plant extract to study the protective effects of the extract against free radicals in 3T3 cells. Some researchers used 0.03%, 0.1%, 0.05% as final concentration of DMSO in cell culture treatment. I want to use 0.4% as final concentration in my work. Is there any problems with this concentration of DMSO in cell viability?

The highest conc. of DMSO you can use is 0.1% but you should try to decrease the conc. as much as you can.
It is good to take into consideration the experimental model that you are using. For example, the time of exposure affects directly the tolerability of your assay. It would be interesting to conduct a cell viability test with different concentrations of DMSO and keep the lowest one posible.
