I do not quite understand about the function of helical domain of PI3Kα. The following paper stated that this domain acts as a scaffold for other domains of p110. I do not have an experience about protein, its structure, its function of each domains, etc. Meanwhile, other papers said that function of this domain was unknown. This domain and kinase domain also have many cases of mutation. If helical domain is mutated, what is the effect?
scaffold domain provide the site for other domain or protein to dock for its proper folding or in a sense it provide a framework for other domain. Generally in a proteins having multiple domain, irrespective of their function known or unknown, all domain play role in the protein function. If there is some critical mutation in its helical domain, it may lead to inactivity of the whole protein or the domain.
