I am presenting an article in weekly journal club meeting and the authors have used " Masson's Trichrome Staining and Picrosirius red staining to asses fibrosis & collagen content, respectively".
Is Masson's trichrome also not used for collagen? I mean collagen as a marker of fibrosis?
I had experience on both of them, and they are both used to stain the collagen fibers, in other words, the results will be almost the same. Sometimes, we just do both of them for double confirm, because many editors think that it is too subjective to conduct the quantification of collagen fibers via histological methods; therefore, collagen assay is also required.

thank you for the answers. I certainly second you both. But, the paper read like they used Masson's for Fibrosis and Picrosirius red for collagen. I was confused as I have always red Masson's stain collagen which is actually a marker for fibrosis.

For example, I usually focus on the liver fibrosis, liver fibrosis is characterized by an excessive accumulation of extracellular matrix (ECM), such as collagen, secreted by myofibroblast-like hepatic stellate cells (also called activated HSCs) mainly.
According to the description above, Fibrosis = excessive fibers = Collagen accumulation, therefore, both of MT & PSRed are common histological methods for COLLAGEN and FIBERS. Of course, both of them are as known as fibrosis marker (exactly, this term indicates fibers too).
Both Masson and Picrosirius Red are staining collagen in histologic preparations. But, differently from aniline blue (the Masson trichromic method dye linking to basic molecular sites of collagenic molecules), Picrosirius Red enhances the intrinsic birefringence of collagen fibres, easily demonstrable at the polarizing microscope (both linear, and (better) with circular polarizing microscopy). I think both of them are the same in heart study for assessing fibrosis.
The different contributions are pointing towards the same marker which is fibrosis. I quite agree with Alberto Calligaro about the scientific difference between the two methods. However, for double confirmation purpose and to avoid subjective bias, both methods are necessary.
