I am using spirit blue agar to screen for lipases in my phage-based metagenomic library. This is proving problematic and I would like to go back to the basics, however, nowhere online can I find an explanation as to how exactly this assay works! What chemical reaction is it that causes the dye to precipitate? I am aware of the process of lipolysis but where does the dye come into it?

Can anyone help?
