I am looking for best methods that can tell me quantitatively about 1.Has antibody bound to GNP and (2.) If yes, then how much of it. The purpose is to ascertain the surface coverage of antibodies on GNP.

From my point of view, acidic hydrolysis and amino acid analysis is the most reliable method. This is a very good method to determine the amount of protein. However, it will not tell you, how much of your antibody is still active on the particles or the selective antigen binding capacity of your gold nanoparticles. Perhaps you want to know these numbers and not 1. +2.?

you can quantitatively determine GNPs linked to the antibody by ICP-OES and ICP-MS after dialyses of GNPs-antibody or filtering
