I was having a discussion with a friend about the best way to resuspend a cell pellet, flicking the tube or pippeting up and down. I wanted to bring the discussion to this community and ask what is your preferred method, does it depend on the cell type, and why would one way be better than the other.

If you are talking about eukaryotic cells and you are concerned about keeping them intact and alive, you should use a gentle method, such as swirling, very gentle vortexing, or gentle pipetting with a 1-ml tip. Avoid methods that introduce bubbles or cause foaming.
If you are talking about bacterial cells, you can use more vigorous methods.
