I'm looking for a way to enable a direct measurement of amino acid content in cell culture media using LCMS.
Many literature use C18 or HILIC column and analyse the sample in +ve mode. However, I think the high salt content in the cell media does cause a lot of trouble in ESI +ve mode. But if I analyse in -ve mode some of the amino acids do not fly.
So what would be the best way to analyse the majority of essential amino acid in cell culture media without a complex sample processing procedure? What's the preferable mobile phase for either C18 or HILIC column?

Mobile phases ACN/0.1%formic acid (A) and water/0.1%formic acid (B).
