I would like to make a homogeneous sterile Pythium suspension to use in plate assays to test whether certain bacterial/fungal species limit the growth of Pythium. I want to be able to spread it over PDA and use it in other various diffusion assays.
So far, I have tried to make a suspension by adding either sterile PBS or sterile demi water with 0,5% Tween to a plate with Pythium. I added this suspension to a sterile bottle with grinding beads and vigorously shook it to break down the mycelium and get a homogenous suspension. I looked at the spores underneath the microscope and also plated the suspensions on PDA, but the results were not optimal.
