I have two questions:
1.
I am trying to improve my IHC staining in cerebellar OSC (penetration wise).
Taking some z-sections, I was able to see (different staining/Abs') only around 40 um depth of staining (out of probably around 150 um total slice width).
2.
I am looking for a recommendation of a working Calbinidin and MOG Abs', for those cerebellar OSC.

For how long do you permeabilise your slices and wht is the composition of the blocking buffer?
I use the calbindin antibody from abcam: ab11426, 1in 1000 dilution

Thanks for your answers Anuja,
For permabilisation, I use 1%Triton-x in PBS overnight at 4OC.
I block with 20%BSA and 0.1%Triton-x in PBS 1h at RT or overnight at 4OC.
