I am doing some experiments on virus infectivity. My positive control cells did not support strong virus replication. The cells in culture look very unhealthy after 2 days and I always make sure to use a new culture medium bottle and FBS. When I performed DAPI staining, there was some evidence of mycoplasma contamination and I believe this is affecting my virus infectivity study as well.
Ciproflaxin at low concentration can eliminate mycoplasma contamination. However, efficacy of killing depends on extent of contamination. Mycoplasma detection and elimination kits are available commercially. You can check with Sigma, invivogen, creative-bioarray.
its difficult to eliminate mycoplasma contamination in some cells like IPS (other stem cells) or its depend on the cell types. PSCeasy mycoplasma detection kit work well.
