I would like to detect human papiloma virus (all types) using PCR. I would like to know if there is anyone who has experience and knows what is the best method for detection of hpv in blood.
You can detect HPV DNA in blood samples as the HPV DNA have been already detected in peripheral blood samples by others. Please take a look at the attached articles !
Do the DNA extraction first by precipitation from blood samples ( using RIBO-prep Kit or u can use Qiagen DNeasy blood & tissue kit ) and then the PCR, you can use АmpliSens HPV HCR-genotype and АmpliSens HPV LCR-genotype Kits
All the tests mentioned above have been validated on LC samples, however, you might need to re-establish the sensitivity of the assay in blood, as HPV might not be found in circulation all throughout the infection...hope this helps!!
I have also used the INNOLipa kit for PCR and genotyping and have had good results with that. You should be able to use this in conjunction with a DNA extraction kit such as the Qiagen blood and tissue kit. Obviously you need to take great care when it comes to contamination (separate PCR hood and preferably room etc). The Doorbar lab have extensive experience with this if you need further help.
Detecting HPV from blood? but it is almost impossible. HPV enters to body from skin, mucus,sexual and cause an topical lesion( such wart, neoplasia) and shedding from that topical region too. Intrinsic immunity system clear it and prevent from a circulating infection, so there is not an accurate Ag- Ab system for serological detection. So the best location for detecting HPV infections is the shedding area( affected area), which in most cases are tissues or cells. Therefore detection of HPV from blood is very difficult. Nevertheless the best primer for detecting all HPV types is from L1 gene, a common primer Known with MY 09/11 is good working.
