One way is to massage the caudal of rat and then after erection of penis, use the body of 1 ml syringe with small rubber tube moist with lubricant as artificial vagina.

cut the syring at mark 4 or 5 and then insert small rubber tube about 0.5 cm with lubrecant into the syring and using the base of needle to block the end of syring body.

The other can be use of potable electroejaculator for collection of semen. But that device is costly nearly 1300 pounds

Actually, for the semen collection without scarifying the rats is the caudal massage using artificial vagina (mentioned earlier by our friend Ayyaz). I have used this method and it's easy but you need assistance to help you and you need to training the animal for that at least 2 weeks before you strat the experiment. Briefly, lay the rats on his back with smooth whole message starting from the head to the tail ( 3-4 times), then start caudal message at the base of the penis in one direction from back to front until the penis erect and make sure the animal relaxed and wait till ejaculate normally without use of artificial vagina ( 3-4 times) and then finally after that try to use the artificial vagina and complete your training for at least one week, after that you can collect easily (be careful, patient and caution when you collecting samples this your first time).

I think by cutting epididymis and collection of it in BSA solution.

With high respect to your opinion, this method can be used for one time only and collect semen in PBS better than BSA solution. the second thing this technique needs a surgical operation under general anesthesia or sacrificing the animals.

actually this is my technique in collecting semen from rats without scarifying them. and i did not published it anywhere and same time no one did it before except me as my knowledge. but it is easy and need practice many times. i am sure you will success. i combined between 2 methods first same method for collecting semen from Japanese quails and other one is from rabbit technique.
