Actually I wanted to know what is the PDT of the breast cancer cell lines I am using for my work. That's why I wanted to know about the method to perform the growth curve analysis.

fit an exponential growth curve to the data, using any maths/stats package.
ie N/N0 = e kt where N0 = initial cell population, N = popn at time t , k = growth constant.
then , having estimated k, t doubling is t when N/No = 2 , ie t doub = ln 2/.k

I would base the growth curve on cell counting instead of proliferation assays. MTT is known to be metabolized to different degree in different cell lines.
