If you do as Vandana mentioned above, the dose of UV is difficult to determine.

Currently in our lab we are using the UV chamber which allow you to determine and fix your uv dose according to your requirements.

I have used low dose, UVC irradiation to induce cellular changes in addition to DNA damage. You should be careful in choosing your dose. If you want primarily damage DNA, UVC is the best as it is closest to the absorption spectra of nucleic acids. PMID: 19003995

I am using Uvitec CL-508.G UV cross linker which emits UV light at 254 nm wavelength (UV C). So how much UV and time needed to Introduce Double stranded breaks. I am suppose to monitor DNA damage makers in the cells by WB and RT PCR.
