If I want to extract a large plasmid from bacteria which are more than 30 kb, what is the best kit to extract it?
Not certain if it is the best but I use the Qiagen kit (or similar kit) but when I elute, I heat the elution buffer up to about 45~60ºC. This improves the elution yield and the overall purification.
Qiagen kit will do. It works on plasmids sized from 100~200-kb in our lab (E. coli).
What is the purpose for isolating the plasmids? Will the bacteria be studied, The plasmid, or both? What is the purpose of the study?
Purpose is studying of the plasmids
Gel electrophoresis of total cell lysate (Eckard method) then purify from gel using kit
