If I want to have bacterial samples producing novel biologically active secondary metabolites, what is the best way for selection of the media for cultivation of these bacteria to produce new biologically active secondary metabolites? Please help me if you can, as really I need that urgently.

Mostly appearance of different properties of bacteria depends on media used. As staph auereus shows golden pigment only on nutrient agar of good quality.
Selection of media of desired new unknown target depends on trial and error method
and if the target properties is existing you can search for literature which talks about it
The kind of media used does affect the production of secondary metabolites. So it would be good for you try a couple of different media to start with and then compare the HPLC/TLC profile of the culture broth extracts of each (compare with a standard if you have one). May be you can dig a bit of literature and see what are the most commonly used media for the kind of bacteria you are growing and shortlist a few of them.
You need to see the pathway in which that metabolite fall and try to use the media constitutes which induces that metabolite. Try the different combination of media and check its production at regular interval. Along with it keep in mind, depending on culture stage (eg, log phase, stationary phase etc), temperature etc. condition it may change.good luck

I think that the best way to select the best medium or culture condition for new metabolite synthesis is to realise a comparison of secreted protein by SDS-PAGE. You can realise different culture condition (+/- O2; etc...) and with different medium and compare the profile to a control condition. Then observe in which condition you have an augmention of stripe. Select the condition with the higher number of protein identified.

I think you have to investigate first the type of appropriate culture medium. At first you can use general medium, if the bacterias grow well you can use it. If it does not grow, you have to select another culture medium (may be you have selected a medium with high yeast or starch or something else). Some bacteria also need different type of trace element. Different type of culture condition also will affect to production of secondary metabolite, you also have to investigate the best of speed agitation, time of fermentation and so on, Hope it will help you. sir., i think you can first c the original source of the organisms., for ex if the organism is lignolytic then ur media can be optimised with respect to composition of wood or lignin....this is my suggestion only.... all the best for your work..
