I have very good experience with NucleoSpin Plant II (MARCHEY-NAGEL) or ZR Plant/Seed DNA Miniprep (ZYMO)...

I think both of them you can buy also in plate version..so you can extract 96 samples in the same time..and it's much faster :)

Hi Danial

To my knowledge, there is no kit that can separate the DNAs from nuclei and mitochrondria/chloroplast. You will have to separate the mitochondrias manually and then proceed with DNA/RNA extraction.

so as i understood i need to isolate the mitochondria and then use any DNA and RNA isolation kit which is specified for plants and then proceed to DNA and RNA extraction.

is that right?

in my lab we normally use Qiagen DNA or RNA extraction kit.

is that suitable for this  purpose?

Yes, you will need to isolate nuclei and mitochondria by differential centrifugation after gently breaking the cell (cytosolic) membrane and then use a method of your choice to isolate DNA from two sub cellular fractions.

In order to isolate good quality mitochondrial RNA, avoid delays between steps.

For isolation of RNA, you could either use a method that allows isolation of RNA and DNA both in separate fractions from the same starting material or divide your sample in two portions and isolate the RNA from one and DNA from another. If supply of starting material is not limiting, I prefer the latter approach to get best quality of RNA and DNA.

i believe there is no limitations for materials in the lab so i will isolate the mitochondria and then proceed to DNA and RNA isolation using Qiagen kit
