I want to investigate the influence of certain drugs on the viability of human bone-marrow derived macrophages cultured in vitro. What viability assay do you find to be most suitable for this kind of experiment?
As stated by Malcom, MTT assay is one of the most common types. It should be MTT is not soluble and first needs to be solubilized. XTT on the other had works on the same principle as MTT, but does not require solubilization and is thus quicker.
Also be aware that superoxide can catalyze the reaction in both assays and might interfere with results, so make sure you inhibitors don't increase superoxide.
For Colorimetric based detection, the most commonly used assays are the MTT and XTT where the actively respiring cells (Viable cells) cleave the tetrazolium salts added to the culture medium. The amount of absorbance correlates with viable cell number. Technically, they are also the best assays for measuring cell proliferation in vitro.
