I am trying to induce tissue factor expression on bovine monocytes from whole blood collected in CDPA-1.
Induction using heparinised human blood with LPS is simple, but I am trying to observe tissue factor expression in bovine blood where I am only able to use CDPA-1 anticoagulant. I have tried stimulating the monocytes using a calcium buffer containing the calcium ionophore A23187 and PMA (as used in a paper by Snyder et al, 2007) with little success.
Has anyone stimulated tissue factor in the whole blood of species other than human? Or alternatively, stimulated human blood in non-heparin anticoagulants?
